The voluntary guidance provides a useful conceptual foundation for evaluating gene‑drive organisms; however, its applicability is uneven across organismal categories. While the framework aligns reasonably well with insect‑based and agricultural use cases, it is substantially less suited to microbial systems, which differ in replication dynamics, ecological behavior, horizontal gene transfer, and environmental persistence. As microbial gene‑drive technologies advance, this gap will become increasingly consequential.

A central challenge is the lack of enforceable mechanisms to ensure consistent application of the guidance across Parties. Risk assessment of gene‑drive organisms requires methodological rigor, transparency, and traceability. Without clear enforcement pathways, the guidance risks becoming aspirational rather than operational. This is particularly problematic for organisms capable of rapid spread or global ecological impact.

The current guidance does not sufficiently address how to protect the risk‑assessment process from actors who may intentionally bias, obscure, or manipulate assessments for political, economic, or strategic purposes. Gene‑drive technologies—especially those with potential environmental or public‑health implications—require robust safeguards, including: Independent scientific review panels, Transparent data‑sharing requirements, Conflict‑of‑interest disclosures, and Mechanisms to detect and deter intentional misrepresentation. Strengthening these protections is essential to maintain public trust and scientific integrity.

The guidance appears to be written with insect vectors and agricultural pests as the primary use case. This leaves significant gaps for microbial gene‑drive systems, which present distinct risk‑assessment challenges, including:

  • Rapid evolutionary adaptation

  • High population turnover

  • Horizontal gene transfer

  • Environmental persistence in diverse niches

  • Difficulty in containment and monitoring

  • Potential interactions with human, animal, and environmental microbiomes - particularly with the rise in popularity of probiotics and prebiotics

Given the accelerating development of engineered microbial systems, the absence of microbe‑specific considerations limits the guidance’s relevance and risks under-preparing Parties for the most likely future applications.
